Biologists fear DNA editing procedure can alter human DNA

A group of biologists was alarmed with the use a new genome-editing technique to modify human DNA in a way that it can become hereditary.
The biologists worry that the new technique is so effective and easy to use that some physicians may push ahead with it before its safety can be weigh up. They also want the public to understand the ethical issues surrounding the technique, which could be used to cure genetic diseases, but also to enhance qualities like beauty or intelligence. The latter is a path that many ethicists believe should never be taken.


“You could exert control over human heredity with this technique, and that is why we are raising the issue,” said David Baltimore, a former president of the California Institute of Technology and a member of the group whose paper on the topic was published in the journal Science.

Ethicists have been concerned for decades about the dangers of altering the human germ line — meaning to make changes to human sperm, eggs or embryos that will last through the life of the individual and be passed on to future generations. Until now, these worries have been theoretical. But a technique invented in 2012 makes it possible to edit the genome precisely and with much greater ease. The technique has already been used to edit the genomes of mice, rats and monkeys, and few doubt that it would work the same way in people.

The new genome-editing technique holds the power to repair or enhance any human gene. “It raises the most fundamental of issues about how we are going to view our humanity in the future and whether we are going to take the dramatic step of modifying our own germline and in a sense take control of our genetic destiny, which raises enormous peril for humanity,” said George Daley, a stem cell expert at Boston Children’s Hospital and a member of the group.

The biologists writing in Science support continuing laboratory research with the technique, and few if any scientists believe it is ready for clinical use. Any such use is tightly regulated in the United States and Europe. American scientists, for instance, would have to present a plan to treat genetic diseases in the human germline to the Food and Drug Administration.

The paper’s authors, however, are concerned about countries that have less regulation in science. They urge that “scientists should avoid even attempting, in lax jurisdictions, germ line genome modification for clinical application in humans” until the full implications “are discussed among scientific and governmental organizations.”

Though such a moratorium would not be legally enforceable and might seem unlikely to exert global sway, there is a precedent. In 1975, scientists worldwide were asked to refrain from using a method for manipulating genes, the recombinant DNA technique, until rules had been established.

“We asked at that time that nobody do certain experiments, and in fact nobody did, to my knowledge,” said Baltimore, who was a member of the 1975 group. “So there is a moral authority you can assert from the U.S., and that is what we hope to do.”

Recombinant DNA was the first in a series of ever-improving steps for manipulating genetic material. The chief problem has always been one of accuracy, of editing the DNA at precisely the intended site, since any off-target change could be lethal. Two recent methods, known as zinc fingers and TAL effectors, came close to the goal of accurate genome editing, but both are hard to use. The new genome-editing approach was invented by Jennifer Doudna of the University of California, Berkeley, and Emmanuelle Charpentier of Umea University in Sweden.

Their method, known by the acronym Crispr-Cas9, co-opts the natural immune system with which bacteria remember the DNA of the viruses that attack them so they are ready the next time those same invaders appear. Researchers can simply prime the defense system with a guide sequence of their choice and it will then destroy the matching DNA sequence in any genome presented to it. Doudna is the lead author of the Science article calling for control of the technique and organized the meeting at which the statement was developed.

Though highly efficient, the technique occasionally cuts the genome at unintended sites. The issue of how much mistargeting could be tolerated in a clinical setting is one that Doudna’s group wants to see thoroughly explored before any human genome is edited.

Scientists also say that replacing a defective gene with a normal one may seem entirely harmless but perhaps would not be.
“We worry about people making changes without the knowledge of what those changes mean in terms of the overall genome,” Baltimore said. “I personally think we are just not smart enough — and won’t be for a very long time — to feel comfortable about the consequences of changing heredity, even in a single individual.”
Many ethicists have accepted the idea of gene therapy, changes that die with the patient, but draw a clear line at altering the germline, since these will extend to future generations. The British Parliament in February approved the transfer of mitochondria, small DNA-containing organelles, to human eggs whose own mitochondria are defective. But that technique is less far-reaching because no genes are edited.

There are two broad schools of thought on modifying the human germline, said R. Alta Charo, a bioethicist at the University of Wisconsin and a member of the Doudna group. One is pragmatic and seeks to balance benefit and risk. The other “sets up inherent limits on how much humankind should alter nature,” she said. 
Some Christian doctrines oppose the idea of playing God, whereas in Judaism and Islam there is the notion “that humankind is supposed to improve the world.” She described herself as more of a pragmatist, saying, “I would try to regulate such things rather than shut a new technology down at its beginning.”

Other scientists agree with the Doudna group’s message.
“It is very clear that people will try to do gene editing in humans,” said Rudolf Jaenisch, a stem cell biologist at the Whitehead Institute in Cambridge, Massachusetts, who was not a member of the Doudna group. “This paper calls for a moratorium on any clinical application, which I believe is the right thing to do.”
Writing in Nature last week, Edward Lanphier and other scientists involved in developing the rival zinc finger technique for genome editing also called for a moratorium on human germline modification, saying that use of current technologies would be “dangerous and ethically unacceptable.”

The International Society for Stem Cell Research said Thursday that it supported the proposed moratorium.

The Doudna group calls for public discussion but is also working to develop some more formal process, such as an international meeting convened by the National Academy of Sciences, to establish guidelines for human use of the genome-editing technique.

“We need some principled agreement that we want to enhance humans in this way or we don’t,” Jaenisch said. “You have to have this discussion because people are gearing up to do this.”

Back in April, a team of Chinese construction workers used a 3D printer to construct houses. By day’s end, there were 10 standing. They were compact and fairly bare bones — nothing much to look at besides the “wow!” factor of there being as many as — count them — 10. But this time around, those same builders have taken the wraps off an achievement that’s roundly more impressive.
In Suzhou Industrial Park, adjacent to Shanghai, stands a five-story structure that the WinSun Decoration Design Engineering firm claims is “the world’s tallest 3D-printed building.” Next to it is the equally massive 3D-printed mansion, which measures 11,840 square feet. Like the previous buildings, the walls are comprised of a mix of concrete and recycled waste materials, such as glass and steel, and formed layer by printed layer. The company stated that the total cost for the mansion was roughly $161,000. 
In a broader sense, this latest feat is yet another indication of how rapidly additive manufacturing techniques are advancing. Once used primarily as a means to quickly render miniature model versions of products, the technology has reached a point where large-scale printers are now capable of making life-sized working creations, such as automobiles, in mere days. For instance, it took less than 48 hours for start-up Local Motors to print a two-seater called the Strati into existence and drive it off the showroom.
Many of these designs, however, typically don’t amount to much beyond being passion projects meant to push 3D printing into new frontiers and drum up some publicity along the way. One example of this is the massive 3D Print Canal House that’s being constructed entirely on-site along a canal in Amsterdam, a process that’s slated to take longer and is less feasible than standard construction, Phil Reeves of UK-based 3D printing research firm Econolyst recently told CNN.
More promising, though, is a system developed by Behrokh Khoshnevis, a University of Southern California engineering professor. His concept machine, called Contour Crafting, involves a clever combination of mechanical cranes and 3D layering to print and assemble entire homes simultaneously — complete with insulation and indoor plumbing — in less than a day.

The approach employed by WinSun isn’t anywhere near that level of sophistication, but it may well prove to be the most practical – at least thus far. There is some labor and equipment costs that comes from trucking in and piecing together the various sections on-site, though the manner in which it all comes together is comparable to the ease of prefab assembly. It’s also reportedly greener thanks to the addition of recycled materials. 
To pitch the advantages of their technology, the company held a news conference to announce that they had taken on orders for 20,000 smaller units as well as highlight some significant cost-cutting figures. According toindustry news site 3Der:
The sheer size of the printer allows for a 10x increase in production efficiency. WinSun estimates that 3D printing technology can save between 30 and 60 percent of building materials and shortens production times by 50 to even 70 percent, while decreasing labor costs by 50 up to even 80 percent. Future applications include 3D printed bridges or tall office buildings that can be built right on site.
WinSun did not respond to a request to disclose how they arrived at those numbers, but Enrico Dini, an Italian civil engineer and chairman of competing start-up Monolite, says that he suspects the calculations may be a tad bit inflated. Still, he emphasized that his own data does back up the claim that, compared to conventional methods, layering may boost overall efficiency. 
“It would be very difficult to fabricate such large sections with traditional concrete casting,” he says. “With 3D printing, you have a lot less waste because you’re only printing out as much material as you need and you can custom shape whole sections on the spot, which can be a big challenge.”

One major concern is whether these large-scale dwellings can hold up over time against the elements. According to 3Der, Ma Rongquan, chief engineer of China Construction Bureau, inspected the building’s structural integrity and found them to be up to code, but was careful to note that state officials have yet to establish specific criteria for assessing the long-term safety of 3D printed architecture.   
And as Dini, who supports the technology, points out, there is the possibility that the use of additive manufacturing may pose some degree of risk. “The only issue is that as the layers of concrete are bonded together, they’re drying at slightly different rates and that’s not very ideal,” he explains. “So there’s maybe a higher chance of it fracturing at the contact point if there’s a strong enough force at play.” 
Regardless, Dini says he’d feel completely safe going inside any floor of either building since construction materials used today are likely to contain special additives to enhance strength and resistance. One such formulation, fiber-reinforced Ductal, has been shown in some tests to be 10 times stronger and last twice as long as regular concrete. He stressed that walls should also be tested to ensure that other properties, such as acoustics, ventilation and thermal insulations are on par with existing buildings.
“In Italy, building standards are extremely strict,” he noted. “But I can’t say I can say the same about China.”

Using X-rays, scientists read 2,000 year old scrolls charred by Mount Vesuvius

Talk about reading between the lines! Scientists wielding X-rays say they can, for the first time, read words inside the charred, rolled-up scrolls that survived the catastrophic eruption of Mt. Vesuvius nearly two millenniums ago.

The findings, described in the journal Nature Communications, give hope to researchers who have until now been unable to read these delicate scrolls without serious risk of destroying them.
The scrolls come from a library in Herculaneum, one of several Roman towns that, along with Pompeii, was destroyed when Mt. Vesuvius erupted in AD 79. This library, a small room in a large villa, held hundreds of handwritten papyrus scrolls that had been carbonized from a furnace-like blast of 608-degree-Fahrenheit gas produced by the volcano.

“This rich book collection, consisting principally of Epicurean philosophical texts, is a unique cultural treasure, as it is the only ancient library to survive together with its books,” the study authors wrote. “The texts preserved in these papyri, now mainly stored in the Officina dei Papiri in the National Library of Naples, had been unknown to scholars before the discovery of the Herculaneum library, since they had not been copied and recopied in late Antiquity, the middle ages and Renaissance.”
So researchers have tried every which way to read these rare and valuable scrolls, which could open a singular window into a lost literary past. The problem is, these scrolls are so delicate that it’s nearly impossible to unroll them without harming them. That hasn’t kept other researchers from trying, however – sometimes successfully, and sometimes not.

“Different opening techniques, all less effective, have been tried over the years until the so-called ‘Oslo method’ was applied in the 1980s on two Herculaneum scrolls now in Paris with problematic results, since the method required the rolls to be picked apart into small pieces,” the study authors wrote. Any further attempts to physically open these scrolls were called off since then, they said, “because an excessive percentage of these ancient texts was irretrievably lost by the application of such methods.”
This is where a technique like X-ray computed tomography, which could penetrate the rolled scrolls, would come in handy. The problem is, the ancient writers used ink made of carbon pulled from smoke residue. And because the papyrus had been carbonized from the blazing heat, both paper and ink are made of roughly the same stuff. Because the soot-based ink and baked paper have about the same density, until now it’s been practically impossible to tell ink and paper apart.

But a team led by Vito Mocella of the Institute for Microelectronics and Microsystems in Naples, Italy, realized they could use a different technique called X-ray phase-contrast tomography. Unlike the standard X-ray CT scans, X-ray phase-contrast tomography examines phase shifts in the X-ray light as it passes through different structures.
Using the technique, the scientists were able to make out a few words and letters from two scrolls, one of them still rolled.

Reading these scrolls is difficult; computer reconstructions of the rolled scroll reveal that the blast of volcanic material so damaged its once-perfect whorls that its cross section looks like a half-melted tree-ring pattern. The paper inside has been thoroughly warped, and some of the letters on the paper probably distorted almost beyond recognition.
Nonetheless, the researchers were able to read a number of words and letters, which were about 2 to 3 millimeters in size. On an unrolled fragment of a scroll called “PHerc.Paris. 1,” they were able to make up the words for “would fall” and “would say.” In the twisted, distorted layers of the rolled-up papyrus called “PHerc.Paris. 4,” they could pick out individual letters: alpha, nu, eta, epsilon and others.

The letters in “PHerc.Paris. 4” are also written in a distinctive style with certain decorative flourishes that seemed very similar to a scroll called “PHerc. 1471,” which holds a text written by the Epicurean philosopher Philodemus. The researchers think they were written in the second quarter of the first century BC.


Ultimately, the researchers wrote, this work was a proofof concept to give other researchers a safe and reliable way to explore ancient philosophical works that were until now off-limits to them.

We encourage you to contact local law enforcement agencies because pets left outside in extreme temperatures, especially without food and shelter, are at risk of hypothermia, frostbite and even death. Their owners are at risk of facing criminal charges.

The act of leaving a pet outside without food or adequate shelter often receives less attention than a violent attack against an animal, but neglect is a crime. "Especially in these cold months, it is important for people to bring their pets inside and for others to report neglected animals to law enforcement,” says Ashley Mauceri, HSUS manager for cruelty response, who fields these calls.


One of the most common forms of animal cruelty, cases of animals left outside in dangerous weather are investigated more by police and animal control agencies than any other form of animal abuse. Our most constant companions—dogs and cats—feel the effects of winter weather as much as we do, only they are often cast outside to weather the cold or a storm owing to a misconception that the fur on their backs will insulate them from suffering. Without proper shelter, food and water, these domesticated animals’ chances of survival in frigid temperatures is greatly decreased. Any pet owners who aren't sure what protections their pets need during cold weather can read our cold-weather advice for keeping pets safe.

While views on animal welfare vary from region to region, there are laws in place in every state to prevent needless suffering. Callers to The HSUS report numerous cases across the country of animals left out in the cold, but the organization is also working with an increasing number of law enforcement agencies that recognize the importance of intervention in these cases.

Longer life depends on sense of meaning and purpose

People with the greatest well-being lives longer than people with least well-being according to a new study. The study was based on an eight-year follow-up period of 9,050 English people. The researchers from the University College London (UCL), Princeton University, and Stony Brook University surveyed 9,050 participants with an average age of 65. The questionnaires measured “eudemonic well-being” of the participants. “Eudemonic well-being,” pertains to the individual’s sense of control, feeling that what you do is worthwhile and sense of purpose in life.
According to the researchers, they already adjusted factors that might affect the result of the survey such as demographics, physical activity and alcohol intake.

During the duration of the study, only nine percent of the people with great well-being died while 29 percent in the least well-being category died. With all other factors considered, the result of the study shows that people with the highest well-being were 30 percent less likely to die. It means they live two years longer than people do in the least-well-being group.

Professor Andrew Steptoe, director of the UCL Institute of Epidemiology and Health Care, who led the study, concludes that the result shows ho happiness is associated with risk of death. “These analyses show that the meaningfulness and sense of purpose that older people have in their lives are also related to survival,” he further added.

Visionary technology figure Elon Musk has been warning the public for months about possible threats posed by artificial intelligence. But now he has a timeline.

Musk, the South African-born CEO of Tesla Motors and SpaceX, wrote that “the risk of something seriously dangerous happening is in the five year time frame. 10 years at most.”

Musk penned this comment at the bottom of an essay written by virtual pioneer Jaron Lanier called “The Myth of A.I.” The essay appeared in the publication Edge.org last week and was followed by comments from such technology notables as George Dyson, Peter Diamandis, and Kevin Kelly.

While Musk’s comment was deleted, it was picked up by sites such as Mashable and Reddit, which preserved it for a larger audience. According to these sites, Musk zeroed in on companies such as DeepMind, a British artificial intelligence company that Musk once invested in before it was purchased by Google.

“The pace of progress in artificial intelligence (I’m not referring to narrow AI) is incredibly fast,” Musk’s piece read. “Unless you have direct exposure to groups like DeepMind, you have no idea how fast-it is growing at a pace close to exponential.”

Musk mentioned that AI companies “recognize the danger” and were working to ameliorate any negative intelligences “from escaping into the Internet.”

While Musk had sent his comment privately to Edge.org by email, it was published by a site editor before it was taken down. A Musk spokesperson has said that Musk will write a longer piece outlining his thoughts on the dangers of artificial intelligence, presumably to be published on the same website.

Previously, Musk has compared AI to “summoning the demon,” nuclear war, and the “Terminator” series.

It's been two weeks since the fatal crash of Virgin Galactic's SpaceShipTwo. As authorities continue to investigate what caused the crash, Virgin Galactic says the accident will not hinder the company's plans to send tourists to space.

It has been two weeks since a Virgin Galactic SpaceShipTwo crashed in the Mojave Desert. The crash was a big strike against the company that is trying to create a commercial private space industry.

Virgin Galactic CEO George Whitesides told a crowd Thursday that the company will continue pursuing its mission to take tourists to space. He made the comments while speaking in Culver City, Calif., to a panel organized by the Rand Corporation.

“We have a new spaceship that’s going to be ready in a few months,” Mr. Whitesides said, according to The Los Angeles Times. “So we’re going to make sure we get that one as safe as we can and keep going.”

On Oct. 31, a SpaceShipTwo crashed during a test flight, killing one pilot and injuring another. The debris from the flight was strewn across 35 miles of the Mojave Desert. Whitesides said that was "a tragic day."

Virgin Galactic is the brainchild of British billionaire Richard Branson. He is planning to take tourists to space for $250,000 a piece. After the crash, there have been growing concerns about the companies safety practices. Twenty of the 700 people who have already paid for the trip have asked for a refund after the crash.

On Nov. 7, Peter Siebold, the pilot who survived the crash, told the National Transportation Safety Board what happened shortly before the crash. He said the ship began to break apart 50,000 feet in the air, according to Engadget. He unbuckled from his seat and a parachute automatically deployed. An investigation by the NTSB found that the SpaceShipTwo's feather re-entry system was a possible cause of the crash. The other pilot, Michael Alsbury, unlocked the system earlier than intended. The NTSB said it could be another year before the final conclusion about what caused the crash is known.

The SpaceShipTwo had only conducted a few dozen test flights. And the fatal flight was only the fourth time that the rocket engines fired. The ship was also using new fuel, which engineers thought would give the engines more thrust.

“There’s so many exciting things happening now in the world of space," he said. "There’s risk but there’s great reward. ... So I think being unafraid to try new things is important as we move forward.”

The robotic lander that touched down on a comet on Wednesday came to rest on its side in the shadow of a cliff, according to the first data beamed home from the probe.

Pictures from cameras on board the European Space Agency’s Philae lander show the machine with one foot in the sky and lodged against a high cliff face that is blocking sunlight to its solar panels.
The precarious resting place means mission controllers are faced with some tough decisions over whether to try and nudge the spacecraft into a sunnier spot. If successful, that would allow Philae to fully recharge its batteries and do more science on the comet, but any sudden move could risk toppling the lander over, or worse, knock it off the comet completely.

The washing machine-sized lander was released by its Rosetta mother ship at 0835am GMT on Wednesday morning and touched down at a perfect spot on the comet’s surface. But when anchoring harpoons failed to fire, the probe bounced back off into space. So weak is the gravitational pull of the comet that Philae soared 1km into the sky and did not come down again until two hours later. “We made quite a leap,” said Stephan Ulamec, the Philae lander manager.

In the time it took the probe to land for the second time, the comet had rotated, bringing more treacherous terrain underneath. The spacecraft bounced a second time and finally came to a standstill on its side at what may be the rim of an enormous crater.

“We bounced twice and stopped in a place we’ve not entirely located,” said Jean-Pierre Bibring, Philae’s lead scientist. Teams of scientists are now trying to work out where the probe is. What mission controllers do know is that they are not where they hoped to be. “We are exactly below a cliff, so we are in a shadow permanently,” Bibring added.

With most of Philae in the dark, the lander will receive only a fraction of the solar energy that Esa had hoped for. The spacecraft needs six or seven hours of sunlight a day but is expected to receive just one and a half. Though it can operate for 60 hours on primary batteries, the probe must then switch to its main batteries which need to be recharged through its solar arrays. If Philae’s batteries run out it will go into a hibernation mode until they have more power.

The spacecraft was designed with landing gear that could hop the probe around, but from its awkward position on its side the option is considered too risky.

Though caught in a tight spot, the Philae lander’s systems appear to be working well. The Rosetta spacecraft picked up the lander’s signal on Thursday morning and received the first images and more instrument data from the surface of the comet.

One of Philae’s major scientific goals is to analyse the comet for organic molecules. To do that, the lander must get samples from the comet into several different instruments, named Ptolemy, Cosac and Civa. There are two ways to do this: sniffing and drilling. Sniffing involves opening the instruments to allow molecules from the surface to drift inside. The instruments are already doing this and returning data.

Drilling is much riskier because it could make the lander topple over... Pushing down into the surface will push the lander off again. “We don’t want to start drilling and end the mission,” said Bibring.
But the team has decided to operate another moving instrument, named Mupus, on Thursday evening. This could cause Philae to shift, but calculations show that it would be in a direction that could improve the amount of sunlight falling on the probe. A change in angle of only a few degrees could help. A new panoramic image will be taken after the Mupus deployment to see if there has been any movement.

Meanwhile, the Rosetta orbiter team will continue to try to pinpoint Philae’s position.
